Anna was born in Portland, Maine, the daughter of William K and Janice Kendrick. She is of English, Irish, and Scottish descent. Her older brother is actor Michael Cooke Kendrick. He appeared in the 2000 movie Looking for an Echo. She attended Deering High School in her hometown.

Anna began her career at the age of 10 when she caught a bus from her hometown to New York City where she could attend auditions. Her first role was as Dinah in the Broadway musical High Society in August 1998 when she was only 12 years old, a performance for which she won the Theatre World Award. She was also nominated for a Tony Award and a Drama Desk Award. The Tony Award nomination made her the third-youngest Tony Award nominee ever. Kendrick went on to appear in a number of other stage productions, including the musical A Little Night Music, playing Fredrika Armfeldt in the New York City Opera production. A little later, she made her film debut in the 2003 musical comedy Camp. Due to her portrayal of Fritzi Wagner in this film, she received a nomination for the Independent Spirit Awards Best Debut Performance.

In 2007, Anna appeared in her next film, Rocket Science, in which she portrayed Ginny Ryerson, a high school debater that talked fast. Although she said she found the role intimidating and challenging after watching an actual national collegiate debate, her performance was praised by critics and she was nominated for the Independent Spirit award for Best Supporting Actress. Later that year, she tried out for the part of Jessica Stanley in the film adaptation of Twilight, a 2008 film based on the initial book of the series by Stephenie Meyer. Anna had intended to do auditions with various actors, but she was too ill and had to leave prematurely. Fortunately for her, she was brought in for a later session where she was given the role of Jessica. Kendrick reprised this role in the first three Twilight sequels.

Anna's singing experience in theater gave her the opportunity to land a role in The Marc Pease Experience (2009), where she appears as a high school senior involved in musical theater. She also appeared in the 2009 film Elsewhere, in which she portrays her first lead character, a girl whose best friend goes missing. She also starred in the movie Up in the Air, based on Walter Kirn's 2001 novel with the same name. For her role in Up in the Air, she won Best Suporting Actress with the National Board of Review, and she also picked up nominations from numerous other awards including The Screen Actors Guild, The Golden Globes, and the Academy Awards.

She reprised her role as Jessica Stanley in The Twilight Saga: Eclipse. She was featured in Edgar Wright's adaptation of the graphic novel Scott Pilgrim, titled Scott Pilgrim vs. the World. In this film, she played Stacey Pilgrim, the sister of the lead character. In 2010, she was in LCD Soundsystem's music video "Pow Pow" as a "shape-shifter" who gathers "the souls of wicked men". In October 2010, she was at the 2010 Top Glamour Awards in Mexico where she won the award for "Best International Actress".

In 2011, she starred in the dramatic comedy 50/50, and reprised her role as Jessica Stanley in The Twilight Saga: Breaking Dawn. In 2012, she played Rosie in the screen adaptation of the baby book What to Expect When You're Expecting. She also voiced the character Courtney in the animated film ParaNorman. She starred in Pitch Perfect as Beca, a rebellious student who learns that she has a voice in her school's female a cappella group, and she is also featured on the film's soundtrack. Kendrick also appeared in the police drama End of Watch, portraying Janet. In 2012 she was in the political thriller The Company You Keep.

In 2013, she scored a popular song with "Cups (When I'm Gone)" from the film Pitch Perfect which became a No. 2 hit on Billboard's Adult Pop Song chart. Anna also hosted SNL on April 5, 2014 with musical guest Pharrell.
