She rose to international fame after her performance as Natalie Keener in the movie Up in the Air (2009), for which she received the Academy Awards, BAFTA, Golden Globe, and Screen Actors Guild Awards for Best Supporting Actress. Anna was born in the state of Maine, the daughter of Janice and William K. Kendrick. Her descent is Scottish, English, and Irish. Her older brother is actor Michael Cooke Kendrick, who appeared in the 2000 film Looking for an Echo. Anna attended Deering High School in her hometown.

Kendrick started her performing career at the age of 10 when she caught a bus from her hometown to New York City where she could try out for roles. Kendrick's initial role was as Dinah in the Broadway musical High Society in August 1998 when she was only 12 years old, a performance for which she was awarded the Theatre World Award. Also, she was nominated for a Drama Desk Award and Tony Award. The Tony Award nomination made her the third-youngest Tony Award nominee ever. Anna went on to perform in a number of other stage productions, including the musical A Little Night Music, in which she played Fredrika Armfeldt in the New York City Opera production. Afterwards, she made her film debut in the 2003 musical comedy Camp. For her portrayal of Fritzi Wagner in this film, she was nominated for the Independent Spirit Awards Best Debut Performance.

In 2007, she appeared in her next film, Rocket Science, in which she portrayed Ginny Ryerson, a fast-talking high school debater. Although she said she found the role challenging and intimidating after watching an actual national collegiate debate, her performance was praised by critics and she was nominated for the Independent Spirit award for Best Supporting Actress. Later that year, Anna tried out for the role of Jessica Stanley in the movie Twilight, a 2008 film based on the initial book of the series by Stephenie Meyer. Anna had intended to do auditions with various actors, but was too sick and had to leave prematurely. Fortunately for her, she was able to come back for a later session and was given the role of Jessica. Kendrick reprised this role in the first three Twilight sequels.

Anna's theater singing experience gave her the chance to star in The Marc Pease Experience (2009), where she appears as a high school senior involved in musical theater. She also appeared in the 2009 film Elsewhere, in which she portrays her first lead character, a girl whose best friend disappears. She also appeared in the film Up in the Air, based on Walter Kirn's 2001 novel of the same name. For her role in Up in the Air, the National Board of Review named her Best Supporting Actress, and she also picked up nominations from numerous other awards including The Screen Actors Guild, The Golden Globes, and the Academy Awards.

She reprised her performance as Jessica Stanley in The Twilight Saga: Eclipse. She appeared in Edgar Wright's adaptation of the graphic novel Scott Pilgrim, titled Scott Pilgrim vs. the World. In this film, she played Stacey Pilgrim, the sister of the lead character. In 2010, she appeared in LCD Soundsystem's video "Pow Pow" as a "shape-shifter" who gathers "the souls of wicked men". In October 2010, she attended the 2010 Top Glamour Awards based in Mexico where she was given the award for "Best International Actress".

In 2011, she starred in the dramatic comedy 50/50, and reprised her role as Jessica Stanley in The Twilight Saga: Breaking Dawn. In 2012, she played Rosie in the film version of the baby manual What to Expect When You're Expecting. She also lent her voice to the character Courtney in the animated movie ParaNorman. She starred in Pitch Perfect as Beca, a rebellious college student who discovers her voice in her school's all-women a cappella group, and she is also featured on the film's soundtrack. Kendrick was also in the police drama End of Watch, playing Janet. In 2012 she appeared in the thriller The Company You Keep.

In 2013, she scored a hit song with "Cups (When I'm Gone)" from the movie Pitch Perfect which became a No. 2 hit on Billboard's Adult Pop Song chart. Kendrick also hosted SNL on April 5, 2014 with musical guest Pharrell.
