Kendrick was born in Portland, Maine, the daughter of Janice and William K. Kendrick. Her descent is Scottish, English, and Irish. Her older brother is actor Michael Cooke Kendrick. He appeared in the 2000 movie Looking for an Echo. Anna attended Deering High School in Portland, Maine.

Anna began her career at the early age of 10 when she caught a bus from Portland, Maine, to New York City where she could attend auditions. Her initial acting role was as Dinah in the Broadway musical High Society in August 1998 when she was only 12 years old, a role for which she won the Theatre World Award. Also, she was nominated for a Drama Desk Award and Tony Award. The Tony Award nomination made her the third-youngest Tony Award nominee ever. Kendrick went on to appear in a number of other stage productions, including A Little Night Music, in which she played Fredrika Armfeldt in the New York City Opera production. A little later, she made her motion picture debut in the 2003 comedy musical Camp. For her portrayal of Fritzi Wagner in this film, she was nominated for the Independent Spirit Awards Best Debut Performance.

In 2007, she had a role in the film Rocket Science, in which she played Ginny Ryerson, a fast-talking high school debater. Although Kendrick said she found the part intimidating and challenging after watching a real national collegiate championship debate, her performance was praised by critics and she was nominated for the Independent Spirit award for Best Supporting Actress. Later that year, Anna tried out for the role of Jessica Stanley in the movie Twilight, a 2008 film based on the initial book of the series by Stephenie Meyer. She had intended to do auditions with various actors, but she was too ill and had to leave prematurely. Fortunately for her, she was brought in for a later session where she won the role of Jessica. Kendrick reprised this role in the first three Twilight sequels.

Kendrick's theater singing experience won her the chance to star in The Marc Pease Experience (2009), where she appears as a high school senior involved in musical theater. She also appeared in the 2009 film Elsewhere, in which she portrays her first lead character, a girl whose best friend disappears. She also starred in the movie Up in the Air, based on Walter Kirn's 2001 novel of the same name. For her role in Up in the Air, she won Best Suporting Actress with the National Board of Review, and she also picked up nominations from numerous other awards including The Screen Actors Guild, The Golden Globes, and the Academy Awards.

Kendrick reprised her role as Jessica Stanley in The Twilight Saga: Eclipse. She was featured in Edgar Wright's rendition of the graphic novel Scott Pilgrim, titled Scott Pilgrim vs. the World. In this film, she played Stacey Pilgrim, the sister of the lead character. In 2010, she appeared in LCD Soundsystem's video "Pow Pow" as a "shape-shifter" who gathers "the souls of wicked men". In October 2010, she was at the 2010 Top Glamour Awards in Mexico where she won the award for "Best International Actress".

In 2011, she starred in the dramatic comedy 50/50, and reprised her her portrayal of Jessica Stanley in The Twilight Saga: Breaking Dawn. In 2012, she was Rosie in the screen adaptation of the baby book What to Expect When You're Expecting. She also voiced the character Courtney in the stop-motion animated movie ParaNorman. She starred in Pitch Perfect as Beca, a rebellious college student who discovers her voice in her school's all-women a cappella group, and she is also featured on the film's soundtrack. Kendrick was also in the police drama End of Watch, portraying Janet. In 2012 she was in the political thriller The Company You Keep.

In 2013, she scored a popular song with "Cups (When I'm Gone)" from the film Pitch Perfect which became a No. 2 hit on Billboard's Adult Pop Song chart. Anna also hosted SNL on April 5, 2014 with musical guest Pharrell.
